How It Works
Reticulocytes are immature red blood cells that retain ribosomal RNA and are identified by new methylene blue staining. Their presence in peripheral blood indicates active erythropoiesis (bone marrow red cell production). In dogs, reticulocytes are classified as aggregate (large, clumped RNA — newly released, counted for regeneration assessment) and punctate (small dots — older, not clinically significant for regeneration). The corrected reticulocyte percentage adjusts for anaemia severity: Corrected % = (Measured % × PCV) ÷ Normal PCV (45%). The Reticulocyte Production Index (RPI) further adjusts for reticulocyte maturation time. An RPI >1 indicates regenerative response; <1 indicates non-regenerative anaemia.
